چکیده

Chemotaxis is the consequence of environmental factors engaging their receptors to initiate signaling cascades. However, the biochemical mechanisms controlling this phenomenon are not clear. We employed an in vitro modified Boyden 48-well chemotaxis migration system to characterize the role of signal transducers in type IV collagen (CIV) induced A2058 human melanoma cell migration. Using specific pharmacological inhibitors and a series of dominant-negative and constitutively active signaling proteins, we show that Ras and Rac GTPases, PI-3K, and PKC participate in cell migration mediated by b1 integrins. Collagen also induces a timedependent degradation of IjB-a and an increase in nuclear translocation of NF-jB which is dependent on PKC pathway. More importantly, collagen-stimulated melanoma cell migration directly correlated with an increase in NF-jB transactivation. Furthermore, CIV induced an increase in b1 integrin mRNA levels. Specific NF-jB inhibitors Helenalin and SN-50 inhibited melanoma cell migration to collagen, indicating a novel requirement for NF-jB transactivation in cell chemotaxis mediated by b1 integrin signals. These results describe signal transduction events that are initiated by type IV collagen through b1 integrins and demonstrate an important role for NF-jB in regulating melanoma chemotaxis. Type IV collagen-mediated melanoma cell adhesion and migration: involvement of multiple, distinct domains of the collagen molecule.

Tumor cell metastasis involves a complex series of interdependent events, including repeated invasion of basement membranes. Studies from several laboratories have implicated tumor cell adhesion and migration in response to laminin as a major contributing factor in tumor cell invasion. The current studies address the direct role of type IV collagen in promoting tumor cell adhesion, spreading, a...

Collagen IV regulates Caco-2 migration and ERK activation via alpha1beta1- and alpha2beta1-integrin-dependent Src kinase activation.

Our previous work indicates intestinal epithelial cell ERK activation by collagen IV, a major component of the intestinal epithelial basement membrane, requires focal adhesion kinase (FAK) and suggests FAK and ERK may have important roles in regulating intestinal epithelial cell migration. We therefore sought to identify FAK downstream targets regulating intestinal epithelial cell spreading, mi...

The Role of Type IV Collagen in Developing Lens in Mouse Fetuses

Objective(s) Extracellular matrix (ECM) and basement membrane (BM) play important roles in many developmental processes during development and after birth. Among the components of the BM, collagen fibers specially type IV are the most important parts. The aim of this study was to determine the time when collagen type IV appears in the BM of lens structure during mouse embryonic development. M...

Purpose: In this investigation specific antibody type IV collagen has been used in light microscopy to study development of BMG of the embryonic and postnatal mouse glomerular mesangium. Materials and Methods:20 female Balb/C mice were selected randomly and were kept under normal condition, finding vaginal plug was assumed as day zero of pregnancy. 12 pregnant mice were scarified by cervical di...

Characterization of a synthetic peptide from type IV collagen that promotes melanoma cell adhesion, spreading, and motility

The adhesion and motility of tumor cells on basement membranes is a central consideration in tumor cell invasion and metastasis. Basement membrane type IV collagen directly promotes the adhesion and migration of various tumor cell types in vitro. Our previous studies demonstrated that tumor cells adhered and spread on surfaces coated with intact type IV collagen or either of the two major enzym...
